May 21st

Greetings, Friend!

Breakthroughs in neurodegenerative disease treatment are accelerating, with the Alzheimer’s therapy market projected to exceed $15Bn by 2030 and multiple sclerosis treatments expected to surpass $35Bn globally.

A lesser-known, clinical-stage biopharma company is advancing an unconventional immunology-based approach targeting both central nervous system inflammation and disease progression.

Early-stage work suggests potential applications across multiple high-burden indications, positioning the platform within two of heal-thcare’s fastest-growing segments.

The company's differentiated strategy and clinical direction make it a compelling story worth a closer look as the landscape continues to shift.

And with an update analyst target pointing to potential triple-digit upside, this under-the-radar Nasdaq idea tops our watchlist Thursday:

Tiziana Life Sciences, Ltd. (Nasdaq: TLSA)

Tiziana is a clinical-stage biopharmaceutical company developing breakthrough therapies using transformational drug delivery technologies to enable alternative routes of immunotherapy.

The Company's innovative nasal approach has the potential to provide an improvement in efficacy as well as safety and tolerability compared to intravenous (IV) delivery.

Tiziana’s lead candidate, intranasal foralumab, which is the only fully human anti-CD3 mAb currently in clinical development, has demonstrated a favorable safety profile and clinical response in patients in studies to date.

Tiziana Life Sciences is a clinical-stage biopharma company that specializes in the developing transformative therapies for neurodegenerative and neuroinflammatory.

Their clinical pipeline includes drug assets for Secondary Progressive Multiple Sclerosis, Alzheimer’s, and ALS.

Tiziana is led by a team of highly qualified executives with extensive drug development and commercialization experience.

Company Background

Tiziana's mission is to design and deliver next generation immunotherapies for neurodegenerative and neuroinflammatory diseases.

The Company's lead immunotherapeutic candidate, Foralumab (TZLS-401), is being developed for Non-Active Secondary Progressive Multiple Sclerosis, Alzheimer’s and other CNS indications. Foralumab is the only fully human anti-CD3 monoclonal antibody under clinical development and is expected to minimize adverse immune responses in patients.

Tiziana has submitted a patent application on potential use of Foralumab, to improve success of chimeric antigen receptor T cells (CAR-T) therapy for cancer and other human diseases. The patent application covers inventions related to improving CAR-T expansion and/or survival.

The Company employs a lean and virtual research and development model using highly experienced teams of experts for each business function to maximize value accretion by focusing resources on the drug discovery and development processes.

Corporate Strategy

-- Continue the clinical development of intranasal administration of Foralumab for neurodegenerative disease indications:

  • Secondary Progressive, Multiple Sclerosis
  • Alzheimer’s
  • Amyotrophic Lateral Sclerosis (ALS)

-- Commence Phase 2 clinical trial after successfully receiving their Investigational New Drug Application (IND) to conduct a Phase 2 study intranasal foralumab in Alzheimer’s disease patients.

-- Submit an Investigational New Drug Application (IND) to conduct a Phase 2 study intranasal foralumab in Alzheimer’s disease patients.

-- Seek orphan drugs fast track or breakthrough designation for the Company's product candidates where warranted.

Clinical Candidates

Tiziana's major clinical assets are supported by extensive worldwide issued patents and pending patent applications covering composition of matter, formulation technologies, manufacturing processes and disease indications.

Their Lead Asset is Intranasal Foralumab

Multiple administration routes of a proven systemic anti-inflammatory, patent protected until 2040

*Non-Active Secondary Progressive Multiple Sclerosis (expanded program; n=10) ** Grant from Amyotrophic Lateral Sclerosis Association for development

Technology - Foralumab

A Novel Intranasal Immunotherapy to Reduce Neuroinflammation

Intranasal foralumab is successfully delivered to the cervical lymph nodes through a convenient nasal administration.

It binds to the surface of a CD4+ or a CD8+ T Cell.

This stimulates the development of anti-inflammatory producing IL-10 T Regs.

These T Regs migrate into the CNS where they infiltrate the blood brain barrier and produce IL-10 to promote a switch in the activated microglial state to an anti-inflammatory homeostatic state.

Additionally, these IL-10 suppress other cells of the CNS including astrocytes which reduces blood brain barrier degradation

These mechanisms help defend the CNS against chronic inflammatory damage.

Because of this, nasally administered foralumab is a promising therapeutic for numerous neuroinflammatory and neurodegenerative diseases.

5 Key Reasons Why (Nasdaq: TLSA) Holds Our Top Watchlist Spot Thursday

#1. A Newly Upgraded $9 Analyst Target: This week, TLSA earned an upgraded target of $9.00 from Lucid Capital Markets.

From its closing valuation on Wednesday, this target suggests TLSA may have a potential 450+% upside.

#2. Chart Support May Be Strengthening At Key Technical Levels.

TLSA ended Wednesday's session in a noteworthy spot on its chart.

The Nasdaq profile closed above five important technical levels, suggesting that chart support may be gaining momentum.

These 5 technical levels could be building strength:

  • 5-Day Simple Moving Average (SMA)
  • 20-Day SMA
  • 50-Day SMA
  • 5-Day Exponential Moving Average (EMA)
  • 13-Day EMA

Keep an eye on TLSA’s chart, as its 200-Day SMA was tested and momentarily broken through on Wednesday.

#3. Tiziana Announced New Positive Clinical Data for Intranasal Foralumab in Non-Active Secondary Progressive Multiple Sclerosis.

Tiziana Life Sciences reported encouraging updated clinical data from its Expanded Access ("EA") Program evaluating intranasal foralumab in 14 patients with non-active Secondary Progressive Multiple Sclerosis (na-SPMS).

Over extended treatment, the therapy remained highly well tolerated with no new safety concerns.

Patients exhibited stabilization of disability and meaningful improvements in fatigue, key outcomes in a condition with limited options.

These results strengthen confidence in foralumab’s potential as a differentiated, non-invasive treatment and support continued advancement toward regulatory approval and broader clinical adoption.

#4. Tiziana Reported Reduced Brain Inflammation in Multiple System Atrophy Patients Treated with Intranasal Foralumab.

Tiziana Life Sciences announced compelling early Phase 2 data showing intranasal foralumab significantly reduced brain inflammation in two patients with Multiple System Atrophy ("MSA").

PET scans revealed up to 35% reductions in standardized uptake value (SUV) and approximately 24% reduction in standardized uptake value ratio (SUVR) in affected areas of the brain.

Foralumab continues to demonstrate consistent anti-inflammatory benefits across multiple neurological indications.

These findings build on prior success in multiple sclerosis and Alzheimer’s disease, reinforcing the therapy’s broad applicability and positioning Tiziana at the forefront of innovative treatments for neurodegenerative diseases with high unmet need.

#5. Tiziana Life Sciences Announced Publication of Positive Data Demonstrating Intranasal Anti-CD3 Antibody Attenuates Long CVD Neuroinflammation and Improves Cognitive Function.

Tiziana Life Sciences announced the publication of positive preclinical data in a bioRxiv preprint.

The study demonstrated that nasal anti-CD3 treatment significantly reduces neuroinflammation, boosts regulatory T cells (Tregs) in the brain, restores hippocampal neurogenesis, and improves short-term memory in a model of Long CVD.

Independent academic collaboration strengthens credibility and translational relevance, supported by human data linking reduced Tregs to symptoms.

As a non-invasive immunomodulatory approach, foralumab shows potential to address widespread “brain fog.”

These results expand Tiziana’s pipeline opp's and validate its platform across multiple neuroinflammatory conditions, supporting future clinical development initiatives.
